چکیده

Cystinosin is predicted to comprise seven transmembrane domains but its actual topology remains unknown. As a first step to address this issue, we used a classical gene fusion approach in which a topology reporter is fused to various truncated fragments of the membrane protein of interest in order to map the topological orientation of a set of truncation sites along the polypeptide (see Manoil and Beckwith 1986, Science 233:1403-8). A series of C-terminally truncated fragments of human cystinosin linked to a glycosylationbased topology reporter were constructed as depicted in Figure 1.

CRF grant ̈Molecular anatomy and physiology of human cystinosin ̈

As mentioned in the previous report, a series of C-terminally truncated fragments of human cystinosin (pink segment) lacking endogenous N-glycosylation sites was fused to a glycosylation-based topology reporter (in blue) followed by an expression reporter (in green). The glycosylation state of the topology reporter (glycosylated when it is translocated into the ER lumen) indicates the topology ...

Combination of Stem Cell Mobilized by Granulocyte-Colony Stimulating Factor and Human Umbilical Cord Matrix Stem Cell: Therapy of Traumatic Brain Injury in Rats

Objective(s) Clinical studies of treating traumatic brain injury (TBI) with autologous adult stem cells led us to examine the impression of a combination therapy. This was performed by intravenous injection of human umbilical cord matrix stem cell (hUCMSC-Wharton,s jelly stem cell) with bone marrow cell mobilized by granulocytecolony stimulating factor (G-CSF) in rats injured with cortical com...

Identification and subcellular localization of a new cystinosin isoform.

Nephropathic cystinosis is a lysosomal disorder caused by functional defects of cystinosin, which mediates cystine efflux into the cytosol. The protein sequence contains at least two signals that target the protein to the lysosomal compartment, one of which is located at the carboxy terminal tail (GYDQL). We have isolated from a human kidney cDNA library a cystinosin isoform, which is generated...

Comparison of human adipose-derived stem cells and chondroitinase ABC transplantation on locomotor recovery in the contusion model of spinal cord injury in rats

Objective(s):Spinal cord injury (SCI) is one of the most serious clinical diseases and its treatment has been a subject of interest to researchers. There are two important therapeutic strategies in the treatment of SCI: replacing lost tissue cells through cells implantation and scar elimination. Therefore, in this study we used human adipose-derived stem cells (hADSCs) implantation and injectio...

Molecular pathogenesis of cystinosis: effect of CTNS mutations on the transport activity and subcellular localization of cystinosin.

Cystinosis is an inherited disorder characterized by defective lysosomal efflux of cystine. Three clinical forms (infantile, juvenile and ocular cystinosis) have been described according to the age of onset and severity of the symptoms. The causative gene, CTNS, encodes a seven transmembrane domain protein, cystinosin, which we recently identified as a H+-driven cystine transporter using an in ...
